Purification of the COP9 signalosome (CSN) from human erythrocytes. The CSN complex was purified to homogeneity from human blood. (A) Coomassie stained 12% SDS-PAGE describe the CSN eight purified subunits. (B) Immunoblotting described Ub-like protein Nedd8, removed by CSN-mediated deneddyylation.

科学背景

COP9 信号体(CSN)最初在拟南芥中被鉴定为植物光依赖型发育的抑制因子,对植物发育有重要的作用,并且其本身既是激酶的靶标,又与激酶结合并协调其活性。虽然CSN复合物与26S蛋白酶体中19S调节颗粒的“盖子”在结构上具有显著相似性,但它们的功能似乎不同,并且在19S“盖子”中未发现CSN所拥有的蛋白质-蛋白质相互作用或相关活性。CSN似乎是一个26S蛋白酶体的降解过程传递信号的平台。
